Sample Preparation for Solution Digest

Consult with us at pmaf@austin.utexas.edu to determine if your sample is suitable for solution digest. This method will give the highest recovery of peptides, but may not work for high levels of detergent and polymer in the buffer.

We use the SP4 digest protocol which can handle polymers and detergents in moderate amounts.  

Measure the protein concentration with the appropriate assay for your protein and buffer.

Put all the buffer components in the sample submission form. If you have dried your sample, the salts will still be in the sample as well as polymers and detergents. Even MWCO filters will not remove detergents and polymers, so list these in the buffer components.

Put each sample in a separate eppendorf tube.

Fill out a work order for Protein ID on FBS https://fbs.research.utexas.edu/Anon/Logon.aspx   Contact pmaf@austin.utexas.edu if you do not have a log-in yet. The work order will generate a four digit PF#.

Put your order number on the tubes and sequentially number them  (PF#-1,PF#-2,3......)  Bring them to MBB 1.420 or ship them to Proteomics Facility, MBB 1.420, 2500 Speedway Ave, Austin, TX 78712.
